Two question to answer first, 1) where you are and 2) what is your gender.
1) If you live in a third world country such as those in South Caucasus or Middle East, you simple don't respond, because unless it's the tourist next door, no one will answer, it's sort of a taboo and gets revealed after the first night. There is a tiny country called Armenia in South Caucasus, where after the first night, if the girl turns out to be a virgin (and god help her if she does not), the guy's family takes a red apple (symbol for virginity) to the girl's house and all the neighbors see and everyone is happy.
If you are in United States, like me, the question is a pretty ordinary thing to ask, and the answer I think depends on the second question, which is
2) Are you a guy? if the answer is "yes," I'd suggest to go ahead and always answer positively. After all, if you're above 18, what's wrong with you if you still haven't had sex. If you are a girl, I'm not sure, I'm not one, so I haven't considered all the pros and coins of saying yes, but saying "no" is not as easy as saying yes for a guy. (unless she goes through a medical procedure, of course.)
But, like others have suggested, avoid the question if you can do so with a witty response. After all, it's nobody's business and does not say much about the person, on contrary, it allows others to generalize because people need quick ways to judge you without having to go through the hard work of finding out who you really are.
