When you're on stage, make eye contact with the girls who like you. (I know this can be tough if the floodlights are in your eyes, like they were when I was doing open mic comedy.) When you're talking for the next few seconds, do it like you're talking to them. Flirt with that a little, then go back to talking to everyone. Do it a couple times.
Then after the show you can just start talking to them about (how'd they like the show, have they been there before, what is their major, whatever). You already made a connection and you just have to follow up. If there is a group make sure to have a friend -- otherwise do not join a group unless they invite you.
Basic principle: people often break the ice in nonverbal ways. If you and a girl are in the same place at the same time, one of the best ways to get her open to talking to you is by becoming aware that you're both in the same place, looking at the same thing. You look at her, you look away, she looks at you, she looks away. You look at each other. You pause. You talk. Try it next time you're waiting for an elevator
