The important thing is not what you say but how you say it, and this has never been more important than in speechmaking - I've had quite a lot of experience speaking in front of large audiences.
Confidence is not a choice. You have to breathe confidence. As long as you're confident, the audience will be confident enough to enjoy you, confident enough to laugh at your jokes, and confident enough to relax.
How do you get confidence, if you're not already? In this case, preparation. Preparation begets confidence. You should definitely memorize the speech. Then have it to hand just for comfort.
Speak slow, pause, range your tone. Everyone loves you.
It's much like pickup in that to begin with, people will value you by the value you give yourself. If you think you're not a good speechmaker and the audience will not laugh at your jokes, guess what, they won't. And, the opposite is also true.
As for lines, the 'best man' commercial market is relatively huge for such an event, there are a myriad books and info on shit, and frankly, given that you've been chosen as his 'best man', you, more than anyone else, should have the right kind of information to hand that you can use.
