Situation 1 - You are shopping with your target and the salesperson says...."are you two together"

Situation 2 - You are with your target and she keeps asking you what you think of one of her girl friends in a attraction sought of way.

Situation 1: "NO, this is my baby sister, cant you tell?"
this will make her question herself, and might make her qualify herself to you, because she is wondering if you arent attracted to her.

Situation 2: (If the friend actually IS hot), just say "Yes, dont you think so?"
This will intimidate her, and make her think that you are actually just using her as a tool to get to her friend, which will also make her try to qualify herself to you and try to make you see that she is much cooler than her friend. At this point you could do some push-pull.
