if he's going for her just to mess around and not a serious relationship, i'd have some fun with it and have a friendly competition but not let her know the other guy is my buddy.

I've done it quite a few times and it actually helps build the friendship because we would ask each other quiestions on how we succeeded or fucked up and learn from that so that we know how to go about it with a girl we do want as a serious relationship and we r now able to interact.

I think it depends on how strong of a friendship you have with the guy. Close friends? Of course not.

Once I didn't do it, and now regret it.

He is my best friend, he's been with her for 4 years. We talked about it, and I decided not to go for it and made mistake.

In the back of my mind I know that things wouldn't be the same between us, although he said he was ok with that. I like to think that if the roles would different I would give her to him.

Talk to him. This is not about bros and hoes, this is about what you want and being mature enough about this situation.
