I've used DD answer to this:

The girl I used this on is my gf SPAM, great girl. Love this stuff!

i get asked that and my favourite reply is to say

"no"

few seconds pause look them in the eye then say

"its hard to believe isn't it"

you can wink as well for extra cockyness

I don't really like any of the responses you guys posted. What works better is a line from Mystery:

Her: "Do you have a GF?"
Me: "You're attracted to me, aren't you."

You could also use Mystery's kiss gambit:

Her: "Do you have a GF?"
Me: "Do you want to kiss me?"
